Product Introduction
T7 endonuclease I recognizes and cuts incomplete paired DNA, cross-structured DNA, Holliday structured or crossed DNA, heterodouble-stranded DNA or, at a slower rate, double-stranded DNA with incisations.
This enzyme cuts the first, second, or third phosphodiester at the mismatched base 5′ end.
This product is a high purity protein obtained by cloning recombinant expression of T7 endonuclease I gene, which is not contaminated by other endonuclease or exonuclease.
Application
1. Detection of mutants formed by gene mutation, SNP, TALEN or CRISPR/Cas9
2. Identify mismatched DNA
3. Break down the four-way cross DNA or branch DNA
4. Detect or cut heterodimer DNA and cut DNA
5. Randomly cut linear DNA for shoot-gun cloning
Precautions
(1) 1×T7ExoBuffer: 50mMKAc, 20mMTris-Ac, 10mM
Mg(Ac) 2,1 mMDTT (pH7.9 25℃), incubated at 25℃.
(2) The optimal reaction temperature of the enzyme is 25℃, and it can be inactivated for 20min at 75℃.
